چکیده
In psoriasis, keratinocytes of the healthy-looking uninvolved skin are more sensitive to stress. Our previous study suggested that cell-stress proteins showing altered expression in psoriatic vs. healthy can affect mitochondrial functions. Therefore, we aimed examine whether involved these alterations, and compared functions between cultured detail by high–resolution respirometry. We did not find any significant difference oxidative phosphorylation capacity, ATP-independent respiration, residual oxygen consumption keratinocytes. However, maximal respiratory capacity was significantly lower Uninvolved showed a mild increase cytochrome c release cells. H2O2release, as marker reactive species, higher during leak respiration Moreover, observed stronger staining intensity immunolabeling frequent number cells intense conclusion, our results suggest increased biogenesis dysfunctions These alterations sensitivity stress provide further evidence about stressed state epidermis. Further studies may reveal changes have protective role or make susceptible for lesion formation.

Involved and Uninvolved Psoriatic Keratinocytes Display a Resistance to Apoptosis that may Contribute to Epidermal Thickness.
Psoriasis is a common autoimmune skin disease. The aim of this study was to investigate whether the apoptotic process is disturbed in psoriatic keratinocytes. In vitro culture of keratinocytes derived from both involved and uninvolved psoriatic skin, revealed higher viability and resistance to apoptosis following exposure to ultraviolet B, compared with cells from healthy controls. The position...
متن کاملExpression of endoglin in psoriatic involved and uninvolved skin.
Endoglin is a glycoprotein with TGF-beta binding capacity and is predominantly expressed on endothelial cells. In psoriasis, TGF-beta has appeared to play a role in the extravasation of peripheral blood mononuclear cells via the endothelium. In order to find out more about the role of endoglin in psoriasis, immunohistochemical staining with PN-E2, a novel anti-endoglin, and of PAL-E, recognizin...
متن کاملExpression of endoglin in the transition between psoriatic uninvolved and involved skin.
Endoglin is a glycoprotein which is predominantly expressed on endothelial cells. It is upregulated under inflammatory conditions as well as in skin lesions where endothelial cell proliferation occurs. Endoglin has the capacity to bind transforming growth factor beta (TGF-beta) and can reduce the bioavailability of TGF-beta. TGF-beta has a growth-inhibiting effect on keratinocytes and a restrai...
متن کاملAnalysis of epithelial–mesenchymal transition markers in psoriatic epidermal keratinocytes
Psoriasis is similar to endpoints of epithelial-mesenchymal transition (EMT), a process of epithelial cells transformed into fibroblast-like cells. The molecular epithelial and mesenchymal markers were analysed in psoriatic keratinocytes. No obvious alteration of epithelial markers E-cadherin (E-cad), keratin 10 (K10), K14 and K16 was detected in psoriatic keratinocytes. However, significantly ...
متن کاملIncreased DNA synthesis of uninvolved psoriatic epidermis is maintained in vitro.
Clinically uninvolved psoriatic epidermis shows increased DNA synthesis in vivo. We have studied the DNA synthesis of cultured keratinocytes from uninvolved psoriatic skin. Trypsinized epidermal cells were plated on plastic dishes pre-coated with bovine collagen type I. In initial studies, normal human serum was found to be superior to fetal bovine in supporting the growth of human epidermal ke...
